In a mammalian cell, the S phase typically lasts for about 8 hours; in simpler eucaryotic cells such as yeasts, the S phase can be as short as 40 minutes. A working model of DNA replication under cell cycle control has resulted (Figure 1b). However, a fully reconstituted in vitro replication system in which both initiation and elongation occur with all purified components is still lacking. Given all the tools of modern, recombinant DNA technology, this issue should be resolved in the future.

Within eukaryotes, DNA replication is controlled within the context of the cell cycle. As the cell grows and divides, it progresses through stages in the cell cycle; DNA replication takes place during the S phase (synthesis phase). The progress of the eukaryotic cell through the cycle is controlled by cell cycle checkpoints.
